This is a list of all the main story's quests, in the order they are obtained.

Warm Up

Warm Up
I have decided to investigate Hinoe's mountain pass. They call this place 'The Isle of Demons', but the demons themselves appear laughably weak. If they do show their sorry faces, I'd like to give them a good beating. Honestly, I don't know WHY it came to this.
To begin: Reach the Mountain Pass after clearing the Dragon's Tunnel
On completion: Completing this quest is required to continue in the game.
Requirements:

First Rice Crop

First Rice Crop
I cannot entrust that fool samurai with our precious seeds. 'Tis a burdensome task indeed, but it is only fitting that a harvest goddess like myself do the bulk of the planting.
To begin: This is obtained as part of the previous quest, Warm Up; it is received after the conversation with Myrthe, Tauemon and Kinta outside the storehouse.
On completion: The story will progress after completing this quest.

Becoming a Blacksmith

Becoming a Blacksmith
Kinta has taken an interest in the forge. He claims that if I build him a proper smithing shed, he shall craft us tools….Ugh. There's no way I'll get along with that brat, though his offer is certainly tempting.
To begin: Shortly after planting your first rice seedlings, Kinta will be standing outside the house, looking at a sword - talk to him to begin the quest.
On completion: Completing this quest will build Kinta's forging hut and will allow Sakuna to forge new weapons, as well as unlocking all quests relating to Kinta.
Requirements:

Pioneering

Pioneering
My valiant efforts have driven away the nearby demons, making it easier to survive in the wilds. Unfortunately, there's been little to forage. In order to make a decent living, it falls upon me to venture into the lush forests.
To begin: This quest is obtained during mealtime after you clear Eggplant Hollow and complete the main quest Warm Up.

The Second Year's Rice Crop

The Second Year's Rice Crop
The previous year's harvest was wretched, but I will not shrink from my work. If I cannot escape this awful place, then I may as well make myself comfortable for the duration of my exile here.
To begin: This quest automatically begins during the morning of the third part of winter.
On completion: This quest is required to progress in the story.

Kaimaru and the Dog
Kaimaru picked up a dog from…somewhere. But with a canine companion, even the feeble children of men could safely leave the mountain pass. I tire of being the only one working here, so as a test, we shall have Tauemon venture out and gather.

The Weaving Hut of Dreams
Yui's a rather adept weaver! With the proper tools, she could even fashion me some new garb. A charming goddess of my stature shudders at the thought of staying on this island with naught but the clothes on her back...I must craft Yui a weaving hut.
To begin: This quest is obtained after your second rice harvest; Yui can be seen talking to Kaimaru. After the following interaction, Yui will be standing in the middle of the path further down. Talk to her to begin this quest.
On completion: Completing this quest will allow Sakuna to enlist Yui's services to fashion headwear and garments, as well as unlocking all quests relating to Yui.

Kaimaru has up and vanished! What were he and that fool samurai thinking, leaving the mountain pass without a dog at their side? I must hunt them down before anything unfortunate comes of it?

According to Semorigami, the saboteur who mixed something foul into my Revered Ears was none other than Kokorowa. I find that difficult to believe. Whatever the case may be, I must return to the capital, confront my dear friend, and uncover the truth.

The Homusubi specter I fought at the volcano was not entirely vanquished. Now specters are appearing all over the island. I do not know what they are plotting, but I must go face them if I am to protect the children of men and secure our future.

The sword Kinta forged gained a spirit and vanishd off somewhere. I dare not consider what may happen if we let it be, so I must go and search for it. Come to think of it, he did mention a desire to forge swords a while back. And here I took him for a toolmaker.
